The two spotted spider mite, Tetranychus urticae Koch, is a nontarget herbivore of Bt-cotton, but acquires and accumulates higher levels of Cry toxin than that expressed by transgenic plants. This work investigated the development and reproduction of T. urticae and of the predator Phytoseiulus macropilis Banks, during three successive generations looking for potential nontarget effect. In addition, behavioral studies on feeding preference, oviposition, and predation were carried out on Bt and non-Bt cottons. The development and reproduction of T. urticae and P. macropilis was conducted using leaf discs of Bt and non-Bt cottons. Arena containing leaf discs from both cotton types connected by a slide coverslip were also used in the behavioral studies....

Characterization of an Argentine isolate of Bacillus thuringiensis similar to the HD-1 strain Neotropical Entomology
Sauka,Diego H; Basurto-Ríos,Regina E; Ibarra,Jorge E; Benintende,Graciela B.
We report the characterization of an Argentine isolate of Bacillus thuringiensis (INTA TA24-6) similar to the HD-1 strain, which harbors a cryptic cry2Ab gene that apparently is transcribed but not translated into a protein. INTA TA24-6 showed a Rep-PCR pattern identical to the HD-1 strain, a plasmid pattern that resembled that of this strain and cry1 and cry2 genes as HD-1. Screening of cry1 and cry2 genes showed that INTA TA24-6 harbors only cry1Ac and cry2Ab genes. Furthermore, crystalline inclusions of INTA TA24-6 exhibit a bipyramidal shape, typical of Lepidoptera-active B. thuringiensis strains, containing a major protein of ca. 130 kDa toxic to Epinotia aporema Wals. (Lepidoptera: Tortricidae) larvae. Neither the flat-square to cuboidal crystal nor...

Chloroplast localization of Cry1Ac and Cry2A protein- an alternative way of insect control in cotton Biol. Res.
Muzaffar,Adnan; Kiani,Sarfraz; Khan,Muhammad Azmat Ullah; Rao,Abdul Qayyum; Ali,Arfan; Awan,Mudassar Fareed; Iqbal,Adnan; Nasir,Idrees Ahmad; Shahid,Ahmad Ali; Husnain,Tayyab.
BACKGROUND: Insects have developed resistance against Bt-transgenic plants. A multi-barrier defense system to weaken their resistance development is now necessary. One such approach is to use fusion protein genes to increase resistance in plants by introducing more Bt genes in combination. The locating the target protein at the point of insect attack will be more effective. It will not mean that the non-green parts of the plants are free of toxic proteins, but it will inflict more damage on the insects because they are at maximum activity in the green parts of plants. RESULTS: Successful cloning was achieved by the amplification of Cry2A, Cry1Ac, and a transit peptide. The appropriate polymerase chain reaction amplification and digested products confirmed...

EXPRESIÓN DE LA PROTEÍNA Cry1Ac EN TEJIDOS DE LÍNEASTRANSGÉNICAS DE PAPA (Solanum tuberosum spp. ANDÍGENA)VAR. DIACOL CAPIRO. Acta biol.Colomb.

Fitness cost of resistance to Bacillus thuringiensis in velvetbean caterpillar Anticarsia gemmatalis Hübner (Lepidoptera, Noctuidae) Rev. Bras. entomol.
Sosa-Gómez,Daniel Ricardo; Miranda,José Ednilson.
Selection pressure to obtain resistant genotypes can result in fitness cost. In this study, we report the effects of the selection pressure of a commercial formulation of Bacillus thuringiensis on biological aspects of a Dipel-resistant strain of velvetbean caterpillar, Anticarsia gemmatalis Hübner. Comparisons of Dipel-resistant and susceptible individuals revealed significant differences in pupal weight and larval development time. Both strains (Dipel-resistant and susceptible) were susceptible to Cry1Ac toxin expressed in foliar cotton tissues. Resistant and susceptible strains showed low survival rates of 22.5% and 51.2%, respectively, when fed with Greene diet containing Bt-cotton. Larvae bioassayed after three laboratory generations presented lower...

Insect resistance and risk assessment studies of advanced generations of basmati rice expressing two genes of Bacillus thuringiensis Electron. J. Biotechnol.
Rahman,Mahmood-ur; Rashid,Hamza; Shahid,Ahmad Ali; Bashir,Khurram; Husnain,Tayyab; Riazuddin,Sheikh.
Advanced generations of different transgenic lines of indica basmati rice (Basmati-370) expressing two unrelated Bt genes, cry1Ac and cry2A were evaluated for resistance to Yellow Stem Borer (YSB) and Rice Leaf Folder (RLF) under field conditions compared to control lines over three years (2003-2005). Homozygous lines were selected and analyzed for insect resistance, morphological, physiochemical properties and risk assessment studies. After artificial infestation of target insects, the transgenic plants showed significant resistance. Data were recorded in terms of dead hearts and white heads at vegetative and flowering stage respectively. Transgenic lines showed up to 100 and 96% resistance against yellow stem borer at vegetative and flowering stages,...

STUDY OF GENE FLOW FROM GM COTTON (Gossypium hirsutum) VARIETIES IN "EL ESPINAL" (TOLIMA, COLOMBIA) Acta biol.Colomb.
RACHE CARDENAL,LEIDY YANIRA; MORA-OBERLAENDER,JULIAN; CHAPARRO-GIRALDO,ALEJANDRO.
In 2009, 4088 hectares of genetically modified (GM) cotton were planted in Tolima (Colombia), however there is some uncertainty about containment measures needed to prevent the flow of pollen and seed from regulated GM fields into adjacent fields. In this study, the gene flow from GM cotton varieties to conventional or feral cotton plants via seed and pollen was evaluated. ImmunostripTM, PCR and ELISA assays were used to detect gene flow. Fifty six refuges, 27 fields with conventional cotton and four feral individuals of the enterprise "Remolinos Inc." located in El Espinal (Tolima) were analyzed in the first half of 2010. The results indicated seed mediated gene flow in 45 refuges (80.4 %) and 26 fields with conventional cotton (96 %), besides pollen...
